POST: Post Doctoral Fellow(Mechanisms
of Mutagenesis)
A postdoctoral position is
available in the Laboratory of Molecular Genetics at the National Institute of
Environmental Health Sciences, Research Triangle Park, North Carolina, USA. The
laboratory investigates the mechanisms by which organisms produce mutations,
which are being pursued by genetical and biochemical approaches using the
bacterium E. coli as a model system. Specifically, we are aiming to understand
the factors that determine the high-fidelity of chromosomal replication by Pol
III holoenzyme complex (HE). In this project we are studying: (i) mutator
mutants of E. coli whose defect is associated with the polymerase accessory
factors, such as newly isolated mutator mutants of the dnaX gene; (ii) The
role(s) of accessory DNA polymerases in determining DNA replication fidelity;
(iii) the relative contributions of leading and lagging strand DNA replication;
and (iv) the role of deoxynucleoside triphosphate (dNTP) pools in mutagenesis.
A second main area of research is the mechanism of mutagenesis induced by
mutagenic base analogs. Of particular interest is our discovery of a novel
detoxification mechanism for N-hydroxylated base analogs, which has been found
to be dependent on the molybdenum cofactor.
REQUIREMENTS:
Candidates must have a Ph.D.
degree (or equivalent) with less than five years of postdoctoral experience,
and have a demonstrated experience in biochemistry, molecular biology or
genetics.
